Holbrook is a hamlet and census-designated place in Suffolk County, New York, United States. East of Long Island MacArthur Airport sits the community. It has an area of 7.2 sq mi (18.6 km2) without any area of water.
It is surrounded by 6 adjacent hamlets in Suffolk County. Surrounding the north and northeast is Holtsville, North Patchogue in southeast, Sayville and Bayport to the south, Bohemia and Ronkonkoma to the west, and Lake Ronkonkoma in the northwest. The Sunrise Highway interchange or NY 27 which connects the hamlet of Valley Stream from Nassau County and to the easternmost part of the island in Montauk, forms a border separating the Holbrook from the adjacent hamlets to the south. From the interchange Nicolls Rd or CR 97 travels from the south border throughout towards the north intersecting with Long Island Expy interchange just outside the border. Long Island Expy partially passes through the northwest part of the hamlet for only a distance of 0.6 miles (1.12 km). Veterans Memorial Hwy also partially passes through the southwestern area from the west to the Sunrise Highway interchange at the south.
The famous Villa Lombardi's sits at 877 Main St. Adjacent buildings are Seneca Middle School and United States Postal Service. Other attractions around are Holbrook Country Club and Roberts Street Park. Schools within the hamlet include Grundy Avenue Elementary School, Merrimac Elementary School, Seneca Middle School, St John's Lutheran Nursery School and Hiawatha Elementary School in the northwest. The Ronkonkoma Branch of the Long Island Rail Road provides access to Holbrook. Holbrook station closed in 1962, so the nearest access to the line is at Ronkonkoma or Medford stations.
